Pharmacodynamics and pharmacokinetics

Pharmacodynamics

Combined progestogen-estrogen contraceptive drug with a predominant content of gestagen. It inhibits the gonadotropic function of the pituitary gland, blocks gonadotropin, as a result of which the egg does not mature.

Due to the content of estrogen with gestagen, this drug, with certain regimens of use, stimulates the onset of menstruation or delays it. Helps stop uterine bleeding. A higher than optimal dose of estrogen increases the risk of adverse reactions.

Pharmacokinetics

Ethinyl estradiol is absorbed in the small intestine. The maximum concentration in the blood is after 2 hours. Metabolized to hydroxyl derivatives. It is excreted in the form of metabolites by the kidneys (60%), the rest by the intestines.

Norethisterone acetate is completely absorbed. Its maximum concentration in the blood is determined after 0.5-4 hours. Metabolites are also excreted by the kidneys (60%) and intestines (40%).

Impact on a woman’s body

The formula of the medical product is based on a combination of two female hormones. The medicine is a dark green pellet (21 pieces per package). This combination ensures leveling of the function of the pituitary gland (gland of the brain), which controls the functioning of each ovary. In the normal mode of activity of the pituitary gland, it produces hormones that stimulate the ovaries to form the corpus luteum in them and create eggs.

These pills can both delay menstruation and stimulate its onset. It all depends on the application scheme. They can be used:

  • as hormonal contraception;
  • to regulate the timing of the onset of menstruation;
  • to eliminate uterine bleeding;
  • to combat functional or psychological infertility;
  • with uterine hypoplasia (its insufficient development);
  • to eliminate painful menstruation.

This medical product can also help with irregular uterine bleeding following menstrual irregularities.

In this regard, only after examination by a gynecologist, mammologist, blood tests, and blood pressure monitoring can you, with medical assistance, make a decision on the advisability of using Non-Ovlon. A decrease in the contraceptive effect of the drug may be preceded by alcohol abuse and illnesses that will accompany vomiting and diarrhea.

Non-Ovlon, instructions for use (Method and dosage)

Non-Ovlon tablets are taken orally for contraception every evening. Reception begins on the 5th day of menstruation and continues for 21 days. The level of hormones in the blood must be constant, so the interval between doses should be 22-26 hours. At the end of the tablets from one package, take a break of 7 days and begin the next cycle of administration (even if bleeding did not occur during the break).

In the treatment of gynecological diseases, the dose of the drug and dosage regimen are prescribed individually according to indications.

Overdose

The amount of the drug that should enter the body during administration depends on the purposes for which non-ovlon is taken.

Maximum permissible dose

If the described medicine is used to protect against unwanted pregnancy, one tablet per day is enough, starting from the first day of menstruation. This determines the number of dragees in one blister - 21 pieces.

When non-ovlon is taken for medical reasons, the dosage should be determined exclusively by a gynecologist.

Hormonal drug analogues

The German company Schering AG stopped producing the hormonal contraceptive Non-Ovlon due to numerous side effects. But reviews from past years indicate that this drug was reliable, potent and loved by the fair sex. It is for this reason that women and girls who previously took it are looking for analogues.

The hormonal contraceptives listed below are analogs of the drug mentioned and have the same ATC code (international nonproprietary name).

Reviews of Non-Ovlon

This drug belongs to monophasic high-dose oral contraceptives, since the content of ethinyl estradiol in one tablet is 50 mcg. In monophasic preparations, the tablets in the package have a constant composition, so when taking them, a woman will have the same estrogen and gestagen content throughout the entire cycle, which is not physiological. In addition, when taking the drug there is a high risk of side effects, especially thrombotic complications, and cholestasis is often observed. Therefore, recently it has not been used for permanent contraception. According to WHO recommendations, low-dose COCs should be used for this purpose, but some doctors “the old fashioned way” recommend Non-Ovlon.

The drug is widely used for the treatment of gynecological diseases: endometriosis, uterine fibroids, polycystic ovary syndrome, dysfunctional uterine bleeding (DUB). This is evidenced by reviews from patients, which state that the drug was prescribed for therapeutic purposes for glandular cystic endometrial hyperplasia, intermenstrual DUB and juvenile uterine bleeding. The treatment lasted 2-3 months and some women noted weight gain, depressed mood, and pain in the right hypochondrium.

The drug, on the recommendation of a doctor, was also used for emergency contraception during unprotected sexual intercourse. In this case, take 2 tablets no later than 72 hours after intercourse and again 2 tablets after 12 hours. Emergency contraception is an alternative to abortion, but it cannot be used permanently.

Many women take Non-Ovlon when their periods are late and possible pregnancy. Most people prefer not to see a doctor; they forget that taking hormonal drugs that can cause spontaneous abortion can lead to bleeding and other complications, including problems with conception in the future. Take 2 tablets every 12 hours until your period appears, which comes in 1-3 days.

When taking Non-Ovlon if your period is late, you should remember that the method is non-physiological and cannot be used more than 1-2 times a year. After taking the drug for this purpose, the menstrual cycle may be disrupted for a long time, and during subsequent periods the discharge may be more abundant.
