Methionine, one of the active components of the drug, is an essential amino acid that is a source of mobile methyl groups. Methionine is necessary for the synthesis of choline. An increase in choline content helps to increase the synthesis of endogenous (internal) phospholipids and reduce the deposition of neutral fat in the liver. Methionine is also necessary for the neutralization of xenobiotics. In atherosclerosis, it reduces the concentration of cholesterol and increases the concentration of blood phospholipids.
Essential phospholipids are essential factors for the development and functioning of liver cells. The main fraction of essential phospholipids in the preparation is represented by phosphatidylcholine (73%), which is the main component of biological membranes. Once in the body, phosphatidylcholine restores the integrity of the membranes of affected liver cells and activates phospholipid-dependent enzymes located in the membrane, thereby normalizing permeability and enhancing the detoxification and excretory potential of liver cells. Methionine and essential phospholipids enhance the effect of each other, being a source of endogenous (internal) and exogenous (entering the body) phospholipids, respectively, improve the functional state of liver cells and have a hepatoprotective (protective) effect.
Methionine also participates in the metabolism of sulfur-containing amino acids, in the synthesis of epinephrine, creatinine and other biologically active compounds, transforms the action of hormones, vitamins (B12, ascorbic, folic acids), enzymes, proteins, and participates in remethylation, deamination, and decarboxylation reactions. Eslidin restores liver function, increases its detoxification ability, and also normalizes fat, carbon and protein metabolism in the body. Preclinical Safety Data
General toxic effect.
Toxicity of the drug Eslidin® in acute and subchronic experiments. A study of the acute toxicity of the drug Eslidin® was carried out on random-bred white male mice with intraperitoneal administration at doses of 3000, 5000 and 6000 mg/kg. Intoxication (a slight change in behavioral reactions, manifested in the form of increased breathing, grouping of animals, decreased motor activity) in experimental mice developed during the first 30-40 minutes, after which behavioral reactions gradually normalized. No animal deaths were observed. Based on the results of determining acute toxicity when administered into the abdominal cavity of an animal, Eslidin® is classified as toxicity class VI (according to K. K. Sidorov’s classification, it is a relatively harmless substance).
The subchronic toxicity of the drug Eslidin® was studied on 33 random-bred white male rats with an average weight of 206 ± 4 g. The contents of the capsules were administered as an aqueous solution intragastrically once daily, for 1 month at a dose of 70 mg/kg (therapeutic) and 700 mg/kg kg, ten times the therapeutic dose for rats and 70 times the therapeutic dose for humans.
According to the results of the experiment, Eslidin® in the studied doses did not cause significant changes in integral indicators in experimental animals (body weight gain, behavioral reactions, food and water consumption), cellular composition and coagulation parameters of peripheral blood, excretory, absorption, protein-synthesizing, carbohydrate functions of the liver , cholesterol content in blood serum, functional state of the excretory, cardiovascular and nervous systems. Eslidin® had no local irritant effect.
The drug in the studied doses did not cause structural disorders in organs and tissues. Under the influence of the drug at a dose of 700 mg/kg, follicles with wide reactive centers and a significant macrophage reaction were found in the lymph nodes; proliferation of Hassall's bodies was observed in the medulla of the thymus; venous congestion, lymphoid-histiocytic infiltration and mild degeneration of hepatocytes were noted in the liver.
Mutagenicity, carcinogenicity.
In vitro studies with Salmonella and yeast strains, with human cell lines, as well as in vivo studies have not revealed the mutagenic potential of essential phospholipids. According to the FDA (USA), as well as the German Cancer Center, essential phospholipids do not belong to the class of substances with suspected carcinogenicity. No data have been obtained on the carcinogenicity of methionine.
Reproductive toxicity, embryotoxicity, teratogenicity.
No toxic effects were observed in pregnant females and their fetuses when essential phospholipids were administered at doses up to 1000 mg/kg (rats) and 500 mg/kg (rabbits). The administration of essential phospholipids in doses up to 3750 mg/kg orally did not affect perinatal and postnatal development in rats. When essential phospholipids were administered orally at doses of 150, 750 and 3750 mg/kg/day to male (for 10 weeks) and female (2 weeks) rats, no effect on fertility and reproductive ability was detected.
